MMP-9在氧诱导小鼠视网膜新生血管形成中的表达

Expression of MMP-9 in Mice with Oxygen-induced Retinal Neovascularization

摘要: 目的 研究基质金属蛋白酶 9(MMP-9)在氧诱导小鼠视网膜新生血管(RNV)形成中的表达,探讨 MMP-9 抑制剂(GM6001)对 RNV 形成的抑制作用。方法 高氧诱导建立小鼠 RNV 模型。在出氧箱前 1 d(即鼠龄 11 d),分别给予 GM6001 治疗组和高氧对照组小鼠玻璃体腔内注射 GM6001(100 μmol/L)1 μL 和等量 PBS 液,正常对照组和高氧组不做处理。采用 HE 染色方法观察 RNV 情况,采用免疫组织化学、Western blot 和 RT-PCR 法检测 MMP-9 蛋白及基因的表达情况。结果 高氧组和高氧对照组可见大量 RNV 形成,而 GM6001 治疗组 RNV 形成明显减少。与正常对照组相比,高氧组和高氧对照组 MMP-9 蛋白及基因表达水平显著升高,GM6001 治疗组较高氧对照组显著降低,差异均有统计学意义(P < 0.05)。 结论 MMP-9 的异常表达可能与 RNV 形成密切相关,GM6001 能有效抑制 RNV 的形成。

Abstract: Objective To explore the efficacy of GM6001,tissue inhibitor expression and significance of matrix metalloproteinase-9(MMP-9)in mice model of oxygen-induced retinal neovascularization(RNV)and evaluate the inhibition effect of MMP-9 inhibitor(GM6001)on RNV. Methods Mice were placed in oxygen boxes to establish oxygen-induced RNV animal models. The GM6001 treated or hyperxia control groups received an intravitreal injection of 1 μL GM6001(100 μmol/L) or PBS at day 11 after birth. The normal control and hyperxia group were not treated. HE staining was used to detect RNV in retinal whole mounts, the mRNA level and protein expression of MMP-9 were measured by RT-PCR, Western blot and immunohistochemistry, respectively. ResultsRNV in the GM6001 treated group was decreased significantly compared with the hyperxia group and hyperxia control group. Compared with the normal control group, higher protein and mRNA expression of MMP-9 were observed in the hyperxia group and hyperxia control group. The expression of MMP-9 protein and mRNA were decreased in the GM6001 treated group compared with the hyperxia control group(P < 0.05). Conclusion The abnormal expression of MMP-9 was closely correlated with RNV. The development of RNV can be markedly inhibited by MMP-9 inhibitor(GM6001),which, we believe, will provide new molecular targets and therapeutic strategy for retinopathy of prematurity treatment.
