摘要: 目的 实时观测辛二酰苯胺异羟肟酸(SAHA)和肿瘤坏死因子相关凋亡诱导配体(TRAIL)联合使用对三阴性乳腺癌细胞 MDA-MB-231 生长状态的影响。方法 应用实时无标记细胞分析系统(RTCA)动态监测 SAHA 和 TRAIL 联合使用对乳腺癌 MDA-MB-231 细胞生长状况的影响,并通过 BiostationIM 活细胞工作站收集各种处理因素对 MDA-MB-231 细胞增殖干扰作用的 形 态 学 证 据 。 结果 xCELLigence RTCA 显 示 SAHA 和 TRAIL 具 有 协 同 抑 制 MDA - MB - 231 细 胞 生 长 的 作 用 ,其 中 50 ng/mL TRAIL 与 5 μmol/L SAHA 联合作用效果最佳。活细胞工作站显示 SAHA 和 TRAIL 联合处理对 MDA-MB-231 细胞生长的抑制效 果较 SAHA 或 TRAIL 单独处理更显著。结论 SAHA 和 TRAIL 联合使用对三阴性乳腺癌细胞 MDA-MB-231 的生长具有协同抑 制作用。

Abstract: Objective To investigate the effects of combined treatment of suberoylanilide hydroxamic acid(SAHA)and TNF-related apoptosis inducing ligand(TRAIL)on proliferation and morphology change of triple-negative breast cancer cell MDA-MB- 231. Methods The effects of combination treatment of SAHA and TRAIL on proliferation and morphology change of MDA-MB-231 cells were monitored by RTCA. Morphology changes of MDA-MB-231 cells by different treatment factors were observed through time-lapse live cell imaging acquisition. Results Real-time cell proliferation assays showed that a synergistic effect were found when MDA-MB-231 cells were treated with combination of SAHA and TRAIL, and reached the best effect with 5 μmol/L SAHA and 50 ng/mL TRAIL. The Results of time-lapse live cell imaging acquisition showed that the growth inhibition of MDA-MB-231 cells with combined treatment of SAHA and TRAIL were more obvious than that with treatment of SAHA or TRAIL alone. Conclusion The combined treatment of SAHA and TRAIL induces a synergistic effect on growth inhibition in triple - negative breast cancer cell line MDA-MB-231.
