葡萄籽原花青素对肝脏缺血再灌注损伤的保护作用

Protective Effect of Grape Seed Proanthocyanidin against Hepatic Ischemia and Reperfusion Injury

摘要: 目的 研究葡萄籽原花青素对肝脏缺血再灌注损伤的保护作用。方法 建立 C57BL/6 小鼠肝脏缺血再灌注模型。腹 腔注射原花青素,给药量为每天 20 mg/kg,注射 3 周。检测肝脏氧化应激水平以及炎性细胞因子释放水平。结果 原花青素可 降低缺血再灌注导致的谷丙转氨酶及谷草转氨酶活性升高,减少肿瘤坏死因子 α 与白介素的释放,并提高组织的总抗氧化能 力,降低丙二醛水平,从而改善缺血再灌注损伤状况。结论 原花青素可以减少肝脏缺血再灌注的损害,可以作为一种预处理 药物应用于临床。

Abstract: Objective To investigate the protective effect of grape seed proanthocyanidin(GSP)against hepatic ischemia and reperfusion injury(IRI). Methods To build murine warm hepatic IRI model,C57BL/6 mice received 30 min hepatic ischemia process,and then the mice were sacrificed after 1 h reperfusion. The mice were administered with GSP intraperitoneally 20 mg/kg for 3 weeks. Two pathways related to IRI including oxidative stress and pro-inflammatory cytokines release were examined. Results Data showed that GSP could significantly reduce hepatic IRI by decreasing the activity of alanine aminotransferase and aspartate aminotransferase,lessening tumor necrosis factor- α and interleukin-1 expression,enhancing tissue total antioxidative capacity and reducing malondialdehyde level. Conclusion GSP can protect liver against IRI,which can be used as pharmacological preconditioning in clinical practice.
