乙醇通过干预 PGC-1α 表达水平调节 AC16 心肌细胞线粒体生成

Ethanol Modulates Mitochondrial Production of AC16 Cardiomyocytes by Interfering with PGC-1α Expression

摘要: 目的 探究不同浓度乙醇干预下 AC16 心肌细胞过氧化物酶体增殖物激活受体 γ 辅激活因子 1α(PGC-1α)及其下游蛋白核呼吸因子 1(NRF-1)、线粒体转录因子 A(TFAM)表达水平的变化以及心肌细胞线粒体结构功能的改变。方法 应用 MTT 法 检测细胞存活率,流式细胞术检测细胞线粒体膜电位改变,电镜下观察细胞超微结构改变,应用实时定量 PCR、Western blotting 技术分别在基因及蛋白水平检测细胞 PGC-1α、NRF-1、TFAM 表达水平的变化。结果 低浓度乙醇组(50 mmol/L)线粒体数量 增加,线粒体膜电位升高,PGC-1α、NRF-1、TFAM 表达上调;高浓度乙醇组(200 mmol/L)线粒体数量减少、畸变,线粒体膜电位下降, PGC-1α、NRF-1、TFAM 表达下调。结论 乙醇对心血管疾病的影响主要是通过调节心肌细胞中 PGC-1α、NRF-1、TFAM 表达水 平从而影响心肌细胞线粒体生成实现的。

Abstract: Objective To investigate the expression of peroxisome proliferator-activated receptor γ coactivator-1α(PGC-1α),nuclear respiratory factor 1(NRF-1),mitochondrial transcription factor A(TFAM)and mitochondrial structural and functional alterations under different concentrations of ethanol. Methods MTT assay was used to determine the survival rate of AC16 cells under different concentrations of ethanol,and the appropriate concentration of ethanol used for subsequent experiments was selected. Mitochondrial membrane potential of AC16 cells was measured by flow cytometry using fluorescent probe JC- 1. The ultrastructural changes of different groups of cells was observed under the electron microscope. The expressions of PGC-1α and its target proteins NRF-1 and TFAM were detected by real-time PCR and Western blotting. Results The number of mitochondrion,mitochondrial membrane potential,and the expressions of PGC-1α,NRF-1,and TFAM were increased in low-concentration ethanol group(50 mmol/L). While in high-concentration ethanol group(200 mmol/L),they were all decreased,some mitochondria were larger than normal size;the cristae in some mitochondria were disordered,and some mitochondria had no cristae,appearing as circular or elongated vesicles. Conclusion The effect of ethanol on cardiovascular disease is mainly by regulating the expressions of PGC-1α and its downstream proteins NRF-1 and TFAM,which are the key factors in regulating mitochondrial biogenesis.
