益气健脾方对脾气虚证模型大鼠心肌细胞线粒体呼吸链酶 复合物活性的影响

Effect of a Decoction to Nourish Qi and Invigorate the Spleen on the Activity of Mitochondrial Respiratory Chain Enzyme Complexes in Cardiomyocytes of Rats with Spleen Qi Deficiency Syndrome

摘要: 目的 探究益气健脾方对脾气虚证模型大鼠心肌细胞线粒体呼吸链酶复合物活性的影响。方法 将 30 只 SD 大鼠随机 分为正常组、模型组、治疗组,每组 10 只,对 3 组大鼠进行分笼饲养。模型组和治疗组大鼠采取饮食干预联合极限游泳法进行 脾气虚造模,于实验第 15 天对 3 组大鼠的体质量、肛温、食量、饮水量及脾气虚症状积分进行测评。造模成功后,正常组和模型 组采用常规饲养方式联合生理盐水灌胃,治疗组采用常规饲养方式联合益气健脾中药汤剂灌胃,时间为 8 周。之后分别取 3 组 大鼠的心肌线粒体,测定心肌组织线粒体呼吸链酶复合物活性。结果 模型组大鼠的线粒体呼吸链酶复合物Ⅱ和复合物Ⅳ活 性明显低于正常组(P < 0.05);治疗组大鼠的线粒体呼吸链酶复合物Ⅱ和复合物Ⅳ活性与模型组比较差异具有统计学意义(P <0.05)。结论 脾气虚能够造成心肌细胞线粒体发生突变和缺失,导致呼吸链酶复合物活性下降,益气健脾中药能够调节心肌 组织线粒体呼吸链酶复合物Ⅱ和复合物Ⅳ活性。

Abstract: Objective To investigate the effect of a decoction to nourish qi and invigorate the spleen on mitochondrial respiratory chain enzyme complex activity in cardiomyocytes of rats with spleen qi deficiency syndrome. Methods Rats were randomly divided into normal,model,and treatment groups. The model and treatment groups were treated by diet intervention combined with the limit swim method. The general condition and spleen qi deficiency syndrome were assessed on day 15. After the success of the model,the normal and model groups were treated with a conventional feeding method combined with normal saline,and the treatment group was treated by diet intervention combined with a decoction to nourish qi and invigorate the spleen for 9 weeks. The activity of two mitochondrial respiratory chain enzyme complexes was observed. Results The activity of mitochondrial respiratory chain enzyme complexⅡand complex Ⅳ in the model group was significantly lower than the activity in the normal and treatment groups(P < 0.05). The activity levels of complexⅡand complex Ⅳ were significantly different between the model group and the treatment group(P < 0.05). Conclusion Spleen qi deficiency can cause decreased activity of mitochondrial respiratory chain enzyme complexes in myocardial cells. The decoction to nourish qi and invigorate the spleen can modulate the activity of myocardial mitochondrial respiratory chain enzyme complexesⅡand Ⅳ .
