慢性淋巴细胞白血病患者长链非编码RNA RP5-180C18.1的表达及其预后评价作用

Long Non-coding RNA RP5-180C18.1 Expression and Its Prognostic Significance in Patients with Chronic Lymphocytic Leukemia

摘要: 目的 分析慢性淋巴细胞白血病(CLL)患者长链非编码RNA RP5-180C18.1的表达及其在CLL的诊断及预后中的作用。方法 收集本院就诊的CLL患者骨髓液并对患者进行随访,采用qPCR方法检测RP5-180C18.1的相对表达量。结果 与对照组比较,RP5-180C18.1在CLL患者高表达(P=0.013);与IGHV突变型CLL患者比较,RP5-180C18.1在IGHV野生型CLL患者中表达更高(P=0.023);与ZAP70低表达CLL患者比较,RP5-180C18.1在ZAP70高表达CLL患者中表达更高(P=0.002);然而,RP5-180C18.1在CD38高、低表达患者中无统计学差异(P=0.206);RP5-180C18.1高表达CLL患者预后更差,总生存期及无进展生存期更短(P分别为0.048、0.041)。结论 RP5-180C18.1表达可作为CLL诊断及预后分析指标。

Abstract: Objective To investigate the differential expression of RP5-180C18.1 in chronic lymphocytic leukemia (CLL) and to explore its role in the diagnosis and prognosis of CLL. Methods Bone marrow samples of patients with CLL from Shengjing Hospital of China Medical University were collected and analyzed. The relative expression of RP5-180C18.1 was detected by qPCR and analyzed statistically. Results The expression of RP5-180C18.1 in patients with CLL was higher than in healthy controls (P=0.013) and higher in patients with wild-type IGHV CLL (P=0.023) than in those with the IGHV mutation. The expression was also higher in patients with ZAP70 up-regulation CLL (P=0.002) than ZAP70 down-regulation CLL,although no difference was found in patients with CD38 up-regulation CLL compared with CD38 down-regulation CLL. Patients with RP5-180C18.1 up-regulation CLL showed a poor prognosis, including low overall survival rates and low progression-free survival (P=0.048,0.041). Conclusion RP5-180C18.1 can be used for the diagnosis of CLL and for the analysis of its prognosis.
