miR-194对人脑胶质瘤U87细胞恶性生物学行为的影响

Effects of miR-194 on the Malignant Biological Behavior of Human Glioma U87 Cells

摘要: 目的 研究miR-194在人脑胶质瘤组织及人U87细胞中表达,以及其对U87细胞增殖、迁移、侵袭能力和凋亡率的影响。方法 应用实时荧光定量PCR方法分别检测23例人脑胶质瘤组织及5例对照脑组织(脑出血及外伤脑组织)中miR-194的表达水平;将miR-194激动剂、拮抗剂及其各自的空白对照分别转染至U87细胞中,应用CCK-8方法检测转染miR-194后U87细胞增殖能力;Transwell实验检测U87细胞迁移、侵袭能力。流式细胞技术检测转染miR-194后细胞凋亡率。结果 与对照组比较,人脑胶质瘤组织中miR-194表达量显著下调(P<0.05);过表达miR-194后能够明显抑制U87细胞增殖、迁移及侵袭能力,并显著促进U87细胞凋亡。结论 miR-194在胶质瘤组织中表达下调,过表达miR-194能够显著抑制U87细胞增殖、迁移及侵袭能力,促进细胞凋亡。

Abstract: Objective To investigate the expression of miR-194 in human glioma tissues and human U87 cells and to study the effect of miR-194 on the proliferation,migration,invasion,and apoptosis rate of U87 cells. Methods Real-time PCR was used to detect the expression of miR-194 in 23 samples of glioma tissue and 5 samples of control brain tissue (cerebral hemorrhage and traumatic brain tissues). Agomir-194,antagomir-194,and their negative control were transiently transfected into U87 cells. The CCK-8 assay was used to detect the change in proliferative activity of the U87 cells. The Transwell assay was used to detect changes in migration and invasion ability. Flow cytometry was used to detect any changes in cell apoptosis rate after transfection of miR-194. Results Compared with the control group,the expression of miR-194 was significantly down regulated in glioma tissues(P<0.05). Overexpression of miR-194 significantly inhibited the proliferation,migration,and invasion of U87 cells and significantly promoted the apoptosis of U87 cells. Conclusion miR-194 was down regulated in glioma tissues and U87 cells. Overexpression of miR-194 can significantly inhibit the proliferation,migration, and invasion of U87 cells and promote cell apoptosis.
