人脐带干细胞治疗重度烧伤大鼠急性肝损伤的实验研究

Effects of Human Umbilical Cord Stem Cells on Alleviating Acute Hepatic Injury in Severely Burned Rats

摘要: 目的 研究人脐带干细胞治疗重度烧伤大鼠急性肝损伤的效果与机制。方法 将30只SD大鼠随机分为假伤组、烧伤组和干细胞组。烧伤组和干细胞组建立头颈背部35% TBSA Ⅲ度烧伤大鼠模型,采集干细胞移植后24 h的血液和肝组织。免疫组化法检测该干细胞在肝组织中的分布。HE染色检测肝组织的病理学变化。ELISA法检测肝组织的促炎症细胞因子水平。TUNEL染色检测肝细胞的凋亡。血生化自动分析仪检测谷丙转氨酶、谷草转氨酶以及乳酸脱氢酶的水平。对数据进行单因素方差分析和LSD-t检验。结果 人脐带干细胞集中分布于肝组织的小血管内,可显著降低肝组织中肿瘤坏死因子α(TNF-α)、白细胞介素1β(IL-1β)和白细胞介素6(IL-6)的水平、减少肝细胞的凋亡数目,显著减轻大鼠肝组织的病理学改变,改善肝功能,差异有统计学意义(P<0.05)。结论 人脐带干细胞具有保护治疗重度烧伤大鼠急性肝损伤的作用。

Abstract: Objective To investigate the therapeutic effects and mechanisms of action of human umbilical cord stem cells in acute hepatic injury in rats following severe burns. Methods Thirty Sprague-Dawley(SD)rats were randomly allocated to the sham group,the burn group or the stem cell treatment group. Severely burned head,neck,and back of the rat model constituting 35% total burn surface area(TBSA)of Ⅲ degree was confirmed in the burn and stem cell treatment groups. Blood and hepatic tissue samples were collected 24 hours after stem cell transplantation. The stem cell distribution patterns in the hepatic tissues were analyzed by immunohistochemical staining;the levels of TNF-α,IL-1β,and IL-6 by ELISA;hepatocyte apoptosis by TUNEL staining;pathological changes in hepatic tissues by HE staining;and the ALT,AST,and LHD levels in serum using a biochemical blood autoanalyzer. Results The human umbilical cord stem cells were concentrated predominantly in liver capillaries. The stem cell transplantation significantly decreased the levels of proinflammatory factors and hepatocyte apoptosis;as well as markedly alleviated structural damages in the hepatic tissues,thereby improving the liver function in severely burned rats. Conclusion Human umbilical cord stem cells can effectively treat severe burn-induced acute hepatic injuries in rats.
