锶对脂肪干细胞成骨分化中血管内皮生长因子及Notch1信号通路相关蛋白表达的影响

Effects of Strontium on the Expression of VEGF and Notch1 Signaling Pathway in Osteogenic Differentiation of Adipose-derived Stem Cells

摘要: 目的 探究锶(Sr)上调脂肪干细胞(ADSCs)成骨分化过程中血管内皮生长因子(VEGF)的表达与Notch1信号通路的关系。方法 分别用氯化锶和γ-分泌酶抑制剂(DAPT)干预成骨分化诱导的ADSCs,运用Western blotting方法检测Notch1信号通路蛋白(NICD)及VEGF的蛋白表达。结果 与空白对照组相比,Sr组的NICD蛋白表达水平显著升高,DAPT组的NICD蛋白表达水平显著降低;与Sr组相比,Sr+DAPT组的NICD蛋白表达显著降低。与空白对照组相比,Sr组的VEGF蛋白表达水平显著升高,DAPT组的VEGF蛋白表达水平显著降低;与Sr组相比,Sr+DAPT组的VEGF蛋白表达显著降低。结论 Sr能上调ADSCs成骨分化过程中VEGF表达。在Sr促ADSCs成骨分化过程中,Notch1信号通路被激活,并对VEGF的表达起正向调控作用。

Abstract: Objective To investigate the relationship between Notch1 signaling pathway and vascular endothelial growth factor(VEGF) in strontium(Sr)-induced adipose-derived stem cell(ADSC) osteogenic differentiation. Methods The osteogenically induced ADSCs were treated with strontium and γ-secretase inhibitors(DAPT). Western blotting method was used to detect the protein expression level of Notch1 signaling pathway related protein (NICD) and VEGF. Results The expression level of NICD was significantly higher in the Sr-treated group and lower in the DAPT-treated group than in the control group. The expression level of NICD was significantly lower in the Sr and DAPT-treated group than in the Sr-treated group. The expression level of VEGF was significantly higher in the Sr-treated group and lower in the DAPT-treated group than in the control group. The expression level of VEGF was significantly lower in the Sr and DAPT-treated group than in the Sr-treated group. Conclusion Strontium can upregulate the expression level of VEGF in osteogenically induced ADSCs. Notch1 signaling pathway was activated during the strontium-induced osteogenic differentiation of ADSCs,and plays a positive role in regulating VEGF expression.
