兔血清对氧磷酶1对敌敌畏中毒大鼠大脑组织的保护作用

Protective Effect of Serum Paraoxonase 1 on the Brain Tissue of Dichlorvos-poisoned Rats

摘要: 目的 探讨兔血清对氧磷酶1(PON1)对敌敌畏(DDVP)中毒大鼠大脑组织的保护作用。方法 将30只大鼠随机分为5组,对照组予腹腔注射敌敌畏生理盐水1 mL/kg。染毒组予15 mg/kg DDVP腹腔注射。PON1干预组于造模前30 min尾静脉注射PON1 9 600 U/kg,30 min后予15 mg/kg DDVP腹腔注射。传统治疗组予15 mg/kg DDVP腹腔注射后立即予阿托品10 mg/kg+碘解磷定45 mg/kg腹腔注射。联合治疗组于造模前30 min尾静脉注射PON1 9 600 U/kg,30 min后予15 mg/kg DDVP腹腔注射,之后立即予阿托品10 mg/kg+碘解磷定45 mg/kg腹腔注射。各组于造模后8 h处死,运用发光法检测血清中胆碱酯酶(ChE)和过氧化氢酶(CAT)的活性,运用Western blotting观察超氧化物歧化酶1(Sod1)、PTEN诱导激酶1(Pink1)及过氧化物还原酶6(Prdx6)在大脑皮层和大脑海马区中的表达,运用HE染色方法观察其病理改变。结果 血清ChE和CAT检测值说明各组中毒程度,Sod1、Pink1和Prdx6的表达量证明PON1对大脑皮层和海马区均具有保护作用。HE染色可印证上述结果。结论 兔血清PON1对DDVP中毒大鼠大脑组织具有保护作用。

Abstract: Objective To study the protective effect of rabbit serum on the brain tissue of DDVP. Methods Thirty rats were randomly divided into five groups and received the following treatments by intravenous injection(unless otherwise specified):control group, 1 mL/kg dichlorvos in saline; poisoning group, 15 mg/kg DDVP; PON1-intervention group, 9 600 U/kg PON1 into the tail vein 30 min before the molds were made, followed by 15 mg/kg DDVP 30 min later; conventional treatment group, 15 mg/kg DDVP followed by atropine 10 mg/kg + pralidoxime iodide 45 mg/kg; combination treatment group, PON1 9 600 U/kg into the tail vein 30 min before the molds were made, followed by 15 mg/kg DDVP after 30 min and injection of atropine 10 mg/kg + pralidoxime iodide 45 mg/kg into the abdominal cavity immediately afterward. The rats were executed 8 h after the molds were made, and serum ChE and CAT activities were detected by light detection. The protein expression of Sod1, Pink1, and Prdx6 in the cerebral cortex and hippocampus were determined using Western blotting. The pathological changes were observed by using HE staining. Results Serum ChE and CAT indicated the degree of poisoning in each group, and the expressions of Sod1, Pink1, and Prdx6 proved that PON1 exerted protective effects on the cerebral cortex and hippocampal area. These results were verified by analysis of the HE staining. Conclusion Rabbit serum PON1 exerted protective effects on the brain tissue of DDVP intoxicated rats.
