AEG-1和CHD5在胃癌中的表达及其临床意义

AEG-1 and CHD5 Expressions in Gastric Cancer and Their Clinical Significance

摘要: 目的 检测AEG-1和CHD5在胃癌组织及癌旁组织中的表达,探讨两者的表达与胃癌临床病理特征和预后之间的关系。方法 采用免疫组化方法检测58例胃癌组织及其癌旁组织中AEG-1和CHD5的表达情况。结果 AEG-1在胃癌组织中高表达(43/58,74.14%),CHD5在癌旁组织中高表达(41/58,70.69%)。AEG-1的表达与患者的TNM分期、肿瘤浸润深度、淋巴结转移和Lauren分型密切相关(P<0.05);CHD5的表达与患者的肿瘤分化程度、TNM分期和淋巴结转移密切相关(P<0.05)。AEG-1与CHD5蛋白表达水平呈现显著的负相关(r=-0.595,P<0.001)。生存分析显示,AEG-1高表达组5年生存率显著低于低表达组,而CHD5高表达组5年生存率显著高于低表达组。多因素分析显示,TNM分期、肿瘤分化程度、AEG-1与CHD5表达情况是影响胃癌患者预后的独立危险因素。结论 AEG-1和CHD5与胃癌的侵袭和转移关系密切,可能为胃癌患者提供新的预测指标和治疗靶点。

Abstract: Objective To assess AEG-1 and CHD5 expressions in gastric cancer and adjacent tissues and explore the correlation between AEG-1 expression and its clinicopathological characteristics and prognosis in patients with gastric cancer. Methods Immunohistochemistry was performed to assess AEG-1 and CHD5 expressions in 58 gastric cancer tissues and their adjacent tissues. Results AEG-1 is highly expressed in gastric cancer tissues(43/58, 74.14%), whereas CHD5 is highly expressed in adjacent tissues(41/58, 70.69%). AEG-1 expression was closely related to the TNM stage, depth of tumor invasion, lymph node metastasis, and Lauren's classification(P<0.05). CHD5 expression was closely related to the degree of differentiation, TNM stage, and lymph node metastasis(P<0.05). The expression level of AEG-1 protein was significantly and negatively correlated with that of CHD5 protein(r=-0.595, P<0.001). Survival analysis showed that the 5-year survival rate of the AEG-1 overexpression group was significantly lower than that of the AEG-1 low-expression group, whereas the 5-year survival rate of the CHD5 overexpression group was higher than that of the CHD5 low-expression group. Multivariate Cox regression analysis revealed that the TNM stage, tumor differentiation, AEG-1 expression, and CHD5 expression were independent prognostic risk factors in patients with gastric cancer. Conclusion AEG-1 and CHD5 are closely related to the invasion and metastasis of gastric cancer, which provide a new prognostic indicator for survival and therapeutic targets in patients with gastric cancer.
