脱氢表雄酮联合高脂饮食诱导大鼠多囊卵巢综合征模型的研究

Effects of Dehydroepiandrosterone and High-fat Diet on Induction of a Rat Model of Polycystic Ovary Syndrome

摘要: 目的 通过比较脱氢表雄酮(DHEA)联合高脂饮食及单纯DHEA构建多囊卵巢综合征(PCOS)的大鼠模型,探究更理想的多囊卵巢大鼠的建模方法。方法 选取80只21日龄SD雌性大鼠,随机分为对照组、实验组和高脂饮食组3组,其中对照组及实验组大鼠各30只,高脂饮食组大鼠20只。从建模前后大鼠体质量增长、卵巢变化和血清性激素水平等指标进行模型评价。结果 实验组及高脂饮食组血清雄激素水平较对照组均有显著增高,符合多囊卵巢的临床表现。2种造模方法的实验组大鼠均表现出多囊样改变,囊性扩张的卵泡增多,其卵泡内卵母细胞消失,卵巢颗粒细胞层数减少至2~3层,囊状扩张卵泡数量比例明显增多。高脂饮食组大鼠卵巢多囊样改变及卵巢病理学变化较实验组更加显著。结论 DHEA联合高脂饮食构建大鼠模型是更加理想的PCOS动物造模方法。

Abstract: Objective To investigate the effects of DHEA and DHEA associated with a high-fat diet on a rat model of PCOS. Methods Eighty female,3-week-old SD rats were randomly allocated to one of three groups:control group (n=30),experimental group (n=30),and high-fat diet (HFD) group (n=20). After treatment for 8 weeks,the serum concentration of TESTO was determined. The pathological changes in the ovary were examined,and the body weight of the rats was measured every 3 days. Results The serum concentrations of TESTO in the experimental group and the HFD group were higher than those in the control group. More cystic follicles and thicker theca cells,together with fewer layers of granulosa cells,were observed in the experimental group and the HFD group. However,unlike in the experimental group,the changes in the HFD group were more obvious. Conclusion The combination of DHEA and an HFD can lead to the successful induction of a typical PCOS model in rats.
