X-STR侧翼序列SNPs在特殊亲缘关系及混合样品鉴定中的意义

Significance of Using SNPs Located in the X-STR Flanking Region for the Identification of Kinships and Mixed Samples

摘要: 目的 通过STR侧翼序列单核苷酸多态性(SNPs)的筛选,构建解决特殊亲缘关系及二人混合样品的个人识别鉴定的新方法。方法 筛选STR侧翼序列中的SNPs,对包含SNPs的片段进行扩增并测序获得其分型结果,设计序列特异性引物进行扩增并测序。结果 获得该STR位点不同来源个体的分型结果。结论 成功构建基于STR侧翼序列SNPs的二人混合样品的检测方法,并可应用于特殊的亲缘关系鉴定中。

Abstract: Objective A new method was developed for the identification of specific kinships and two-person mixed samples through the screening of X-STR flanking-region single nucleotide polymorphisms (SNPs). Methods The STR flanking region was screened for SNPs. The fragments containing SNPs were then amplified using sequence-specific primers and sequenced for STR locus typing. Results The typing results of the STR locus from different individuals were obtained. Conclusion A detection method based on STR flanking region SNPs was successfully developed for mixed samples containing DNA from two individuals and could be applicable for the identification of kinship.
