Effect of GDNF on COX-2 Expresssion in Rat Hippocampus after Focal Cerebral Ischemia

摘要:

目的观察胶质细胞源性神经营养因子(GDNF)对局灶性脑缺血再灌注损伤(CIPI)大鼠海马环氧酶2(COX-2)表达的影响。方法大脑中动脉线栓法制作大鼠局灶性脑缺血再灌注模型,立体定位下侧脑室注射GDNF,观察模型对照组、生理盐水组、GDNF组大鼠海马COX-2表达。结果GDNF组海马COX-2表达明显减少,与模型对照组、生理盐水组比较差异显著(P<0.05)。结论GDNF可能通过减少海马COX-2表达,增强对局灶性缺血脑组织的保护能力,加速中枢神经损伤的修复。

Abstract:

Objective To observe the effect of glial cell-lined derived
      neurotrophic factor(GDNF)on cyclooxygenase-2(COX-2)expression after focal
      cerebral ischemia/reperfusion injury.Methods The rat model of middle
      cerebral artery occlusion(MACO)was established by intraluminal suture
      method.GDNF was injected into the lateral ventricle under stereo
      localization.The expression of COX-2 in the hippocampus was detected in
      model group,control group,and GDNF group.Results The expression of COX-2
      was significantly lower in GDNF group than in model group and control
      group(P < 0.05).Conclusion GDNF may protect brain tissue from ischemic
      injury by decreasing of the expression of COX-2.
