血必净对高糖环境下大鼠腹膜间皮细胞分泌晚期炎性因子HMGB-1和TGF-β_1的影响

Peritoneal Mesothelial Cells under the Condition of High Glucose

摘要:

目的研究血必净注射液对高糖环境下大鼠腹膜间皮细胞(PMC)分泌高迁移率族蛋白1(HMGB-1)和转化生长因子β1(TGF-β1)的影响,探讨其对腹膜透析相关性腹膜纤维化的防治意义。方法胰蛋白酶消化法行PMC的原代培养和传代,经鉴定第3代细胞用于实验。随机分实验组和对照组,实验组加2.5%的葡萄糖及不同浓度的血必净,培养不同时点分别收集上清液,用ELISA法检测其中HMGB-1和TGF-β1的含量,并进行统计学分析。结果加2.5%葡萄糖的各实验组HMGB-1及TGF-β1的含量均显著高于对照组(P<0.05);加血必净的各实验组HMGB-1及TGF-β1的含量均显著低于单纯高糖组(P<0.05),且随着血必净浓度升高,HMGB-1及TGF-β1的含量下降,20mg/ml血必净组两者含量最低。结论高浓度葡萄糖可促进PMC表达HMGB-1和TGF-β1,而血必净可以拮抗高糖对PMC中HMGB-1和TGF-β1表达的上调作用,提示血必净对预防、延缓腹膜纤维化有一定意义。

Abstract:

Objective In this study,we investigated the effect of Xuebijing on
      the secretion of high mobility group box-1(HMGB-1)and transforming growth
      factor-β1(TGF-β1)in rat peritoneal mesothelial cells(PMC)under the
      condition of high glucose concentration,trying to understand the
      significance of Xuebijing on the prevention and treating of peritoneal
      fibrosis caused by peritoneal dialysis. Methods PMC were isolated from rat
      colic omentum,and the thirdly cultured PMC were used in the experiment.
      PMC were divided into experimental groups and control group at random. The
      experimental groups were treated with 2.5% glucose and Xuebijing of
      different concentrations. The super- nate was collected after 18 h,24 h
      and 36 h to detect HMGB-1 and TGF-β1 in it. Results The expression of
      HMGB-1 and TGF-β1 in all the experimental groups treated with 2.5% glucose
      was significantly higher than that in the control group (P < 0.05).The
      expression of HMGB-1 and TGF-β1 in groups treated with 2.5% glucose and
      Xuebijing was significantly lower than that in groups treated with
      2.5% glucose only (P < 0.05);Xuebijing reduced the synthesis of HMGB-1 and
      TGF-β1 in a dose-dependent manner. Conclusion Glucose of high
      concentration can promote the synthesis of HMGB-1 and TGF-β1 in PMC.
      Xuebijing can reverse these changes and it may have significance in the
      prevention and treating of peritoneal fibrosis.
