胰岛素抵抗大鼠脂肪组织chemerin mRNA的表达和意义

Expressions and Significance of Chemerin mRNA in Adipose Tissue of Insulin Resistant Rats

摘要: 目的 探讨胰岛素抵抗大鼠脂肪组织中脂肪因子chemerin mRNA的表达情况,探讨其与胰岛素抵抗之间的关系及意义。 方法 采用高脂饲料喂养建立胰岛素抵抗大鼠模型,将30只雄性SD大鼠随机分为正常饮食组(NC,n=15)和高脂饮食组(HF,n=15),喂养10周后,以大鼠清醒状态下正常血糖-高血浆胰岛素钳夹技术进行评估,处死后取肾周脂肪组织,应用Real-time PCR方法检测大鼠脂肪组织中chemerin mRNA的表达水平。结果 (1)与NC组比较,HF组60~120 min的平均葡萄糖输注率(GIR60~120)明显降低[(13.87±1.45) mg·kg-1·min-1 vs (24.10±2.87) mg·kg-1·min-1,P<0.05]。(2)与NC组比较,HF组大鼠脂肪组织中chemerin mRNA的表达明显升高[(1.86±1.02) vs (0.92±0.32),P<0.01]。结论 胰岛素抵抗大鼠脂肪组织chemerin mRNA升高,提示脂肪因子chemerin可能参与胰岛素抵抗的发生。

Abstract: Objective To investigate the expressions of chemerin mRNA in adipose tissue of insulin resistant rats and the relationship between them. Methods Insulin resistant rats were induced by high-fat diet feeding. Thirty male SD rats were randomly divided into two groups fed with either basic chow (NC, n=15) or high-fat diet (HF, n=15) for 10 weeks,then assessed by euglycemic-hyperinsulinemia clamp technique. Rats were sacrificed and taken the perinephrit adipose tissue.The chemerin mRNA expression in adipose tissue was tested by real-time PCR. Results (1) The glucose infusion rate(GIR60~120) was significantly decreased in HF group compared with control group, GIR60~120 [(13.87±1.45) mg·kg-1·min-1 vs (24.10±2.87) mg·kg-1·min-1,P<0.05]. (2)The chemerin mRNA expression in adipose tissue was significantly increased in HF group compared with control group [(1.86±1.02) vs (0.92±0.32),P<0.01]. Conclusion The increased expression of chemerin mRNA in adipose tissue of insulin resistant rats showed that chemerin might be involved in insulin resistance.
