血管内皮生长因子D在肌肉瘤小鼠模型中的表达

Expression of Vascular Endothelial Growth Factor D in Animal Models of Myosarcoma

摘要: 目的 建立横纹肌肉瘤模型,研究肌肉瘤淋巴道转移机制,为临床肌肉瘤治疗提供理论依据。方法 取清洁级昆明系小鼠30只,将S180小鼠癌性腹水在小鼠臀部肌层种植,建立肌纤维肉瘤的原位瘤种植模型。观察肿瘤的生长情况,1周后处死小鼠,切取瘤体,检测肿瘤体积、质量,开胸、腹观察肿瘤转移情况,分析不同时期肿瘤血管、淋巴管生长与肿瘤生长转移的关系。取肿瘤组织包埋制备切片,应用免疫组化、Western blot等方法检测血管内皮生长因子D(VEGF-D)的蛋白表达。应用淋巴管内皮透明质酸受体1标记淋巴管,检测肌肉瘤组织中的淋巴管密度。结果 横纹肌肉瘤组织中心区和边缘区均存在淋巴管,其数量随肿瘤的生长明显增加,预示横纹肌肉瘤组织中存在淋巴管的增生和新生,肌肉瘤组织中淋巴管的增生和新生可能与VEGF-D的诱导密切相关。结论 VEGF-D是重要的促淋巴管形成因子,通过增加淋巴管的渗透性,使癌细胞更易进入淋巴管,促进了横纹肌肉瘤的淋巴道转移。

Abstract: Objective To establish rhabdomyosarcoma mouse models for researching the mechanism of lymphatic and hematogenous metastasis of rhabdomyosarcoma. To provide theoretical basis for clinical myosarcoma treatment. Methods Malignant ascites of S180 mouses were planted in muscular layer of gluteal region for establish myofibrosarcoma orthotopic tumor model in 30 sanitation NH mouses. The growth condition of tumor was observed closely. The mouses was killed after the model was set up one week later. The tumors was desected and the volume and weight of tumors were detected. The chest and abdomen were explored to observe condition of neoplasm metastasis, the relation of tumor vessels and the growth of lymphatic invasion and tumor metastatic were analyzed. the tissue of tumor was embeded and sliced for pathological analysis. Immunohistochemisty and Western blot were used to detect the expression of vascular endothelial growth factor D (VEGF-D). LMVD in myosarcoma was calculated by using lymphatic vessel endothelial hyaluronan receptor-1 as the specific marker of lymphatic endothelium. Results There were many lymphatic vessels in centre and marginal zone of rhabdomyosarcoma tissue observed. It showed that lymphatic hyperplasia and neogenesis was seen in rhabdomyosarcoma tissue. Conclusion The positive expression of VEGF-D in rhabdomyosarcoma was related with lymphatic neogenesis, VEGF-D might play an important role and promote lymphatic metastasis of rhabdomyosarcoma.
