乙酰肝素酶与GA结合蛋白α在乳腺癌组织中的表达及意义

摘要: 目的 分析正常乳腺组织和浸润性导管癌中乙酰肝素酶(HPA)和GA结合蛋白α(GABPα)的表达及临床意义,探讨其在乳腺癌发生发展中的作用及相关性。方法 免疫组织化学方法检测HPA及GABPα的表达,并分析二者的相关性及与临床病理因素的关系;Western blot检测HPA和GABPα蛋白的表达。结果 免疫组化显示HPA和GABPα在浸润性导管癌中阳性表达率明显高于正常乳腺组织(P<0.05),且HPA和GABPα的表达与乳腺癌组织学分级、TNM分期、及淋巴结有无转移有关(P<0.05)。Western blot检测显示HPA和GABPα蛋白表达在浸润性导管癌中明显高于正常乳腺组织(P<0.05),与免疫组化的结果一致。统计分析两者在乳腺癌中的表达呈正相关(P<0.05)。结论 在乳腺癌组织中,HPA与GABPα的表达均存在异常,提示HPA和GABPα在乳腺癌的发生发展、浸润转移中可能存在重要的协同作用。

Abstract: Objective To analyze the clinicopathological significance of heparanase(HPA) andGA-binding protein α (GABPα) in normal mammary glands and invasive ductal carcinoma (IDC)and to discuss the role and correlation between them during breast carcinogenesis. Methods HPAand GABPα expression were examined by immuneohistochemistry,and the relationship betweenthe HPA and GABPα protein expression and the clinical pathologic features were analyzed. Western blot analysis was conducted to determine the HPA and GABPα expression. Results Immunohistochemistry showed that heparanase and GABPα were distributed in cytoplasm and nuclear of tumor cells,in contrast to their nonexpression in normal breast tissue. GABP-positive cases and HPA-positive cases had a higher positive rate in breast cancer than that of normal breast tissues(P<0.05).As well as in western blot. The expressions of them were related to histological grade, TNM staging and lymph node metastasis. (P<0.05). And the expression of them were positively correlated in breast cancer( P<0.05).Conclusion Our results indicated that both SIAH2 and Bcl-2 are involved in breast cancer and might imply coordination in breast tumorigenesis.
