Dishevelled-1,3影响肺癌细胞侵袭机制的研究

摘要: 目的 散乱蛋白(Dvl)亚家族成员在非小细胞肺癌中均存在过表达,但不同亚型对肺癌细胞侵袭能力的影响和可能的机制尚不清楚。方法 检测转染Dvl-1和Dvl-3后肺癌细胞系T细胞转录因子(T-cell factor,Tcf)转录活性的改变,β-连环蛋白(β-catenin)和p120连环蛋白(p120ctn)的表达,以及对细胞侵袭能力的影响。结果 Dvl-1和Dvl-3过表达后,肺癌细胞中p120ctn蛋白水平均上调,但Dvl-3可明显上调p120ctn mRNA水平。同时,Dvl-1过表达后Tcf 转录活性明显上调,而Dvl-3未明显影响其Tcf 转录活性。应用β-catenin和p120ctn抗体抑制β-catenin和p120ctn蛋白作用后,可分别显著抑制由Dvl-1和Dvl-3增强的细胞侵袭能力。结论 Dvl-1可能主要通过经典Wnt通路中的β-catenin,而Dvl-3则可能主要通过与非经典Wnt通路相关的p120ctn影响肺癌细胞的生物学行为。

Abstract: Abstract Objective To explore the influence of Dishevelled-1 and 3 over-expression on the invasive ability of lung cancer cells and the underlying mechanisms. Methods The changes of β-catenin-dependent Tcf transcriptional activity, β-catenin and p120ctn expression were detected, together with the invasive ability of lung cancer cells after Dvl-1 and Dvl-3 transfection. Results Exogenous expression of Dvl-1 and Dvl-3 both up-regulated the protein level of p120ctn in lung cancer cells, while only Dvl-3 over-expression was accompanied by increasing p120ctn mRNA. However, Dvl-1 over-expression increased Tcf transcriptional activity significantly compared with Dvl-3. Meanwhile, invasion of Dvl-1 and Dvl-3-enhanced cells was inhibited by both β-catenin and p120ctn antibodies. Conclusion The biological behavior of lung cancer cells may be altered by Dvl-1 mainly through β-catenin which is an important molecule in canonical Wnt pathway, while Dvl-3 is mainly through p120ctn which is related to noncanonical Wnt pathway.
