人脐带间充质干细胞体外修复子痫前期重度患者血清损伤后血管内皮细胞内分泌功能的研究

Effects of Human Umbilical Cord Mesenchymal Stem Cells on the Endocrine Function of Vascular Endothelial Cells after Serum Injury in Patients with Severe Preeclampsia in Vitro

摘要: 目的探讨人脐带间充质干细胞(UC-MSCs)在体外对子痫前期重度患者血清损伤后血管内皮细胞内分泌功能的影响。方法将人脐静脉内皮细胞株接种于6孔培养板,进行分组干预,分为无血清正常对照组(N组)、健康孕妇血清干预组(H组)和子痫前期重度患者血清干预组(P组),每组均予UC-MSCs上清干预。采用ELISA法检测UC-MSCs干预前后细胞上清中ET-1和NO的含量。结果UC-MSCs干预前P组内皮细胞中ET-1含量最高,而NO最低;UC-MSCs干预后P组内皮细胞中ET-1含量下降,而NO升高;UC-MSCs干预前后P组ET-1和NO含量的差异均有统计学意义(P<0.05)。结论UC-MSCs在体外可修复子痫前期重度患者血清损害的内皮细胞的内分泌功能。

Abstract: Abstract: Objective To investigate the effects of umbilical cord mesenchymal stem cells (UC-MSCs) on the endocrine function of vascular endothelial cells after serum injury in patients with severe preeclampsia in vitro. Methods Human umbilical vein endothelial cells (HUVEC) were planted in the six-well cluster dishes and incubated with DMEM with different serum: serum-free normal control group (group N), 15% serum healthy pregnant women (group H) and 15% serum patients with severe preeclampsia (group P). All of the three groups were cultured by UC-MSCs. The contents of ET-1 and NO in the solution of HUVEC were detected before and after the intervention of UC-MSCs by ELISA. Results Before the intervention of UC-MSCs, the content of ET-1 in group P was the highest, while that of NO was the lowest. After the intervention of UC-MSCs, the content of ET-1 in group P decreased, but that of NO increased. The differences between the contents of ET-1 and NO in the solution of HUVEC before and after the intervention of UC-MSCs were statistically significant (P<0.05). Conclusion UC-MSCs can repair the endocrine function of vascular endothelial cells after serum injury in patients with severe preeclampsia in vitro.
