胃癌组织中ATBF1蛋白的表达及临床病理意义

Clinicopathological Significance of ATBF1 Expression in Gastric Carcinoma

摘要: 目的 观察胃癌发生和演进过程中ATBF1蛋白表达的作用及相关机制。方法 福尔马林固定、石蜡包埋的胃癌及癌旁黏膜标本制作成组织芯片,制备4种ATBF1抗体(NT440、1-12、D1-120和AT6)后进行免疫组化反应,检测胃癌中ATBF1蛋白表达,并比较其与胃癌临床病理特征和ki-67表达的关系。 结果 胃癌中细胞质NT440和AT6阳性表达率低于癌旁黏膜(P<0.05),但细胞核NT440的表达与其相反(P<0.05)。老年胃癌患者中D1-120 和AT6的阳性表达率高于年轻患者(P<0.05),D1-120 和AT6表达与胃癌淋巴结转移或局部脉管侵袭呈正相关(P<0.05)。值得注意的是,胃癌中1-12和ki-67阳性表达水平呈正相关(P<0.05)。结论 胃癌发生和演进中异常ATBF1表达及核质转位发挥重要作用,ATBF1表达通过调节细胞增殖参与胃癌局部脉管侵袭和淋巴结转移。

Abstract: Objective To observe the role of ATBF1 expression in and explore relevant mechanisms of tumorigenesis and progression of gastric cancer. Methods Formalin-fixed and paraffin-embedded tissues of gastric cancer (n=85) and adjacent non-neoplastic mucosa (NNM n=84) were studied with tissue microarray for ATBF1 expression using 4 antibodies (NT440, 1-12, D1-120 and AT6) by immunohistochemistry. ATBF1 expression was compared with clinicopathological parameters of tumors, including expression of ki-67 protein. Results The positive rates of cytoplasmic NT440 and AT6 expression were lower in gastric cancers than that in the NNMs (P<0.05), while nuclear NT440 expression showed otherwise (P<0.05). There was higher expression of D1-120 and AT6 in elder than younger patients with gastric cancer (P<0.05). D1-120 expression was positively correlated with lymph node metastasis (P<0.05), which was the same for AT6 expression and angiolymphatic invasion (P<0.05). It was worth noting that 1-12 expression of ATBF1 showed close association with the ki-67 expression in gastric cancer (P<0.05). Conclusion Aberrant ATBF1 expression or its translocation from cytoplasm to nuclear might play an important role during gastric carcinogenesis. ATBF1 expression is involved in local invasion and lymph node metastasis of gastric carcinomas, possibly via regulation of cellular proliferation.
