脂联素基因多态性与新疆地区哈萨克族2型糖尿病的相关性研究

摘要: 目的 研究脂联素基因(APM-1)45T/G单核苷酸多态性(SNPs)与新疆地区哈萨克族人2型糖尿病(T2DM)的相关性。方法:采用病例-对照研究方法,应用聚合酶链反应-限制性内切酶片段长度多态性(PCR-RFLP)技术,比较新疆地区哈萨克族人正常糖耐量组(NGT)和T2DM组APM-1 SNP45基因型与等位基因分布频率差异;比较不同基因型血清APM-1水平以及NGT组不同基因型体测与生化指标的差异。结果:NGT组与T2DM组相比较,APM-1 SNP45位点基因型与等位基因频率以及血清APM-1水平差异均无统计学意义(P>0.05);NGT组TG+GG基因型者体重指数(BMI)、腰围(WAIST)、腰臀比(WHR)及血清甘油三酯(TG)、总胆固醇(CH)和低密度脂蛋白(LDL)水平显著高于TT基因型者(P<0.05),高密度脂蛋白(HDL)水平显著低于TT基因型者(P<0.01)。结论:APM-1 SNP45T/G多态位点可能与新疆地区哈萨克族T2DM无相关性。

Abstract: Objective To investigate the association of adiponectin single nucleotide polymorphism (SNP) 45T/G with the prevalence of type 2 diabetes mellitus (T2DM) in Kazaks of the Xinjiang region, China. Methods The case-control study was employed. The polymerase chain reaction–restriction fragment length polymorphism (PCR-RFLP) method was used to determine the distribution of allele and genotype frequency of the SNP45 T/G polymorphism (exon 2) in the adiponectin gene. ELISA was used to determine plasma adiponectin levels. And many anthropometric and biological indices were compared in different genotype carriers. Results The distribution of genotype and allele frequencies of the SNP45 were not statistically different between T2DM and NGT groups (P>0.05). There was also no significant difference in plasma adiponectin concentrations between TT genotype and TG+GG genotype between T2DM and NGT groups (P>0.05). Compared with SNP45 T carriers, in the NGT group, G carriers had higher body mass index, ratio of waist and Hip,waist circumference,triglyceride, low density lipoprotein and total cholesterol (P < 0.05), and lower levels of high-density lipoprotein (P<0.01). Conclusion SNP45 in adiponectin gene is not associated with T2DM in Kazaks population of Xinjiang.
