Rad18打靶小鼠模型遗传稳定性分析

Genetic integrity analysis of Rad18-/- mice

摘要: 目的探讨RAD18在维持遗传稳定性中的作用。方法对Rad18-/-小鼠模型进行生育力检测。观察不同月龄Rad18-/-雄小鼠睾丸质量、结构以及精子发生和形态的变化。结果Rad18-/-小鼠具有生育能力,但随着年龄增加睾丸质量和生育力下降。组织学检测显示Rad18-/-小鼠睾丸的生精小管有正常的精子发生,睾丸特异性tH2B分布正常。但在12月龄时,25%的生精小管中仅残留已分化的生殖细胞,缺失精原细胞,并且附睾中的精子形态无异常。结论含有精原干细胞的精原细胞耗尽导致了Rad18-/-小鼠随年龄增加而生育力下降。

Abstract: Objective To investigate physiological function of RAD18. Methods Fertility of Rad18-/- male mice was determined. Weight and structure of testis, spermatogenesis and morphology of spermatozoa were compared between wild-type (WT) and Rad18-/- mice at different ages by histological examination. Results Fertility and testis weight of Rad18-/- mice decreased with age. Histological examination showed normal spermatogenesis in seminiferous tubules of Rad18-/- testis, and tH2B had normal distribution as well. However, 25% of the tubules at 12 months retained only late differentiated germ cells without spermatogonia, while the spermatozoa had normal morphology. Conclusion The exhaustion of spermatogonia which contains spermatogonial stem cells results in the decline of fertility with age.
