盐酸巴马汀抑制聚甲基丙烯酸甲酯颗粒诱导骨溶解的实验研究

摘要: 目的 观察破骨细胞在聚甲基丙烯酸甲酯(PMMA)颗粒诱导的骨溶解中的反应,以及盐酸巴马汀对破骨细胞的抑制作用。方法 将40只小鼠随机分为4组,构建颅骨模型。A、D组小鼠颅骨表面注射生理盐水,B、C组注射PMMA颗粒悬液。1 d后A、B组小鼠颅骨植入颗粒处局部注射PBS,C、D组注射盐酸巴马汀。14 d后处死小鼠,取出颅骨进行相关检查。结果 B组骨溶解明显,炎性细胞及破骨细胞计数较其余3组增多(P0.05)。骨矿物质密度和骨矿物含量检测结果显示,A、D组高于B、C组(P<0.05),而C组又明显高于B组(P<0.05)。结论 PMMA颗粒可以诱导骨溶解;盐酸巴马汀可抑制破骨细胞的分化成熟,进而抑制PMMA颗粒诱导的骨溶解,对假体无菌性松动有防治作用。

Abstract: Objective To observe the reaction of osteoclasts in the osteolysis -induced by polymethylmethacrylate (PMMA) particles and the inhibition of palmatine chloride to osteoclasts. Methods Forty40 mice were randomly divided into 4 groups to build establish the skull models. Saline was injected to the mice skull surface of group A and group D. And PMMA particles were injected to the mice skull surface of group B and group C. From the second day on, the PBS was offered to the skull models of group A and group B. Palmatine chloride was offerdofferedadministrated to the skull models of group C and group D. After14 Fourteen days later, killed the mice were sacrificed and removed skulls were separated for the experiment. Results The osteolysis of group B is was obvious and the number of inflammatory cells and osteoclasts wasis more than that of the other three groups (P0.05). The detection shows thatshowed that the bone mineral density (BMD) and bone mineral content (BMC) of group A and group D are were higher than that of group B and group C. W, while the BMD and BMC of group C are were higher than that of group B. Conclusion PMMA particles can induce osteolysis; palmatine chloride can inhibit the differentiation and maturation of osteoclasts and inhibit the osteolysis, which has the preventive function to osteolysis -induced by polymethylmethacrylate particles induced osteolysis.
