鹿茸中促神经生长组分的筛选、鉴定及活性评价

Screening, Identification and Activity Evaluation of the Nerve Growth-Promoting Component in Pilose Antler

摘要: 目的 筛选、鉴定鹿茸中的促神经生长组分,并对其生物学活性进行评价。方法 用标准品rhNGF作用于PC12细胞,建立评价神经生长因子(NGF)生物活性的技术平台,筛选、鉴定鲜鹿茸及鹿茸表皮细胞分泌物中促神经生长组分。结果 鲜鹿茸经匀浆、均质及CM-FF离子交换层析的洗脱#x02161;峰中含有促神经生长组分,经Westernblot法确定其含有NGF的#x003b2;亚基,而鹿茸表皮细胞培养分泌物中无此生物学活性。结论 鲜鹿茸中含有NGF类因子,可以促进PC12细胞的增殖,可能是鹿茸益智的药效组分之一。

Abstract: Objective To screen, identify the nerve growth promoting component in pilose antler, and evaluate its biological activity. Methods The technology platform for the evaluation of NGF biological activity was established. PC12 cells were treated with rhNGF, and then the promoting nerve growth component in fresh antler or its epidermal secretions were screened and identified. Results After homogenization and CM-FF ion exchange chromatography, nerve growth promoting component was identified in the elution peak II of the fresh antler;#x003b2; subunit of NGF was confirmed by WB analysis, which was not observed in the secretion of the antler#x02019;s epidermal cell culture . Conclusion The NGF-like factor was found in the fresh antler, it can promote the proliferation of PC12 cells, which could be one of the main active components in pilose antler.
