脂多糖介导的大鼠颈动脉收缩性增强模型的构建

摘要: 目的 观察脂多糖所致的血管外膜损伤对大鼠颈动脉收缩功能及心肌素等因子表达的影响。方法 利用脂多糖(LPS)刺激大鼠颈动脉外膜;采用不同浓度的5-羟色胺诱发血管痉挛;采用HE染色法观察颈动脉病理改变;Westernblot检测颈动脉中心肌素的变化;实时定量PCR法检测颈动脉中miR-145及miR-1的表达。结果 LPS处理后大鼠颈动脉血管中膜明显增厚,平滑肌细胞数量增多、排列紊乱。应用0.2%的5-羟色胺时,模型组动脉血流量下降程度明显大于对照组。模型组心肌素蛋白的表达水平明显高于对照组(P<0.05),miR-145和miR-1的表达水平明显低于对照组(P<0.05)。结论 本研究应用LPS刺激大鼠颈动脉血管外膜第5天动脉中膜平滑肌增生,并对5-羟色胺的收缩反应增强。模型组血管平滑肌中心肌素的蛋白表达上调,miR-145和miR-1的表达下调。

Abstract: Objective To investigate the effects of lipopolysaccharide (LPS)-mediated adventitial injury on the vasoconstriction of rat carotid artery and the expression of myocardin. Methods Adventitia of rat carotid artery was stimulated using LPS; different concentrations of serotonin was used to induce vasoconstriction; pathological alterations of carotid artery were observed by HE staining; Western blot was performed to measure the expression of myocardin; real-time PCR was adopted to measure the expression of miR-145 and miR-1. Results Microscopic examination demonstrated that media of carotid arteries appeared thicker, smooth muscle cells were increased and disorganized by LPS administration to adventitia. 2#x02030; serotonin-induced reduction of carotid blood flow was significantly augmented in LPS-treated rat compared with that in normal saline-treated rat. Myocardin was significantly upregulated, while miR-145 and miR-1 were significantly reduced in LPS-treated rats. Conclusion 5 days after LPS-stimulated adventitia of rat carotid artery, media smooth muscle cells were increased and hypersensitive to serotonin. Myocardin expression was up-regulated, while miR-145 and miR-1 were down-regulated in LPS-stimulated carotid artery.
