丙型肝炎患者外周血单核细胞中IFNAR2mRNA表达的研究

摘要: 目的探究丙型肝炎病毒(HCV)不同感染阶段,外周血单核细胞(PBMC)干扰素#x003b1;/#x003b2;受体2(IFN-#x003b1;/#x003b2;AR2)mRNA表达水平,并研究其与干扰素(IFN)疗效的关系。方法58名丙肝抗体阳性患者,根据临床表现分为3组:急性肝炎组(4名),慢性肝炎组(46名),肝硬化组(8名)。15名健康志愿者作为对照组。应用密度梯度离心法将外周血单核细胞分离,应用RT-PCR扩增IFNAR2mRNA。结果58名丙型肝炎患中,PBMCIFNAR2mRNA检出率为87.9%),而对照组为33.3%,差异具有统计学意义(P<0.05);慢性丙型肝炎组较肝硬化组检出率高(93.5%#x02236;50%,P<0.05),PBMCHCV-RNA的阳性率无差别(P>0.05)。PBMCIFNAR2mRNA水平与干扰素疗效相关(P<0.05)。结论丙型肝炎病毒感染可上调PBMCIFNAR2mRNA的表达,慢性丙型肝炎患者PBMCIFNAR2mRNA表达高于肝硬化患者,其表达可能与病毒载量无关,而与干扰素疗效相关。

Abstract: Objective To investigate the differences in interferon-#x003b1;/#x003b2; receptor 2 (IFNAR2) mRNA expression level in peripheral blood mononuclear cells (PBMC) at different stages of hepatitis C virus (HCV) infection,and determine whether it has any correlation with the effectiveness of interferon therapy. Methods A total of 58 patients,positive for anti-HCV antibodies,were recruited for the study and divided into three groups depending on their clinical symptoms: acute hepatitis (4 cases),chronic hepatitis (46 cases) and liver cirrhosis (8 cases). 15 volunteers served as healthy controls. PBMC were purified by density gradient centrifugation and IFNAR2 mRNA was amplified by a reverse transcription-polymerase chain reaction (RT-PCR) assay. Results The detection rate of IFNAR2 mRNA was 87.9% (51/58) in the PBMC of HCV patients,which was significantly higher than the control group (33.3%,P<0.05).
