类风湿关节炎患者干扰素调节因子4基因表达变化的研究

IRF4 Expression in Patients with Rheumatoid Arthritis

摘要: 目的通过检测类风湿关节炎(RA)人群和正常对照人群干扰素调节因子4(IRF4)基因表达的差异,探讨其在RA发病中的意义。方法选择30例RA患者与28名正常对照者,取外周血抽提总RNA并反转录为cDNA,采用实时荧光定量聚合酶链反应扩增IRF4基因,计算RA组和正常对照组的IRF4mRNA表达水平的差异(以一#x025B3;#x025B3;CT值表示),并与血沉、C-反应蛋白(CRP)、类风湿因子(RF)、抗环瓜氨酸多肽(抗CCP)抗体水平等指标进行相关性分析。结果RA组、对照组外周血IRF4mRNA的表达水平分别为-6.94#x000b1;O.10,-7.62#x000b1;0.13,RA患者外周血IRF4mRNA的表达水平较正常对照组明显增高(P<0.01),IRF4mRNA水平与疾病活动性指标呈明显相关性改变。结论RA患者外周血存在IRF4表达异常,且与疾病活动指标具有相关性,可能在RA发病中起一定作用。

Abstract: To compare the expression of interferon regulatory factor 4(IRF4) in rheumatoid arthritis(RA) patients and normal individuals,and investigate its role in the development of RA.Methods The clinical data of 30 RA patients and 28 normal controls were collected. Total RNA of peripheral blood was extracted and transcripted into cDNA. Sybr green dye based real time quantitative PCR method was performed to compare the expression (indicated as一#x025B3;#x025B3;CT value) of IRF4 in patients with RA and controls.The correlation of levels of IRF4 mRNA with active index between patients was analyzed. Results The levels of IRF4 mRNA in rheumatoid arthritis patients and normal controls were -6.94#x000b1;O.10 and -7.62#x000b1;0.13,respectively(PIRF4 mRNA were obviously correlated with clinical active index .Conclusion Abnormal IRF4 mRNA expression was found in heperipheral blood of RA patients and associated with clinical active indexs,IRF4 may play a role in the pathogensis of RA development.
