应用导入HSVtk基因的骨髓间充质干细胞治疗胶质瘤的实验研究

In vitro Anti-tumor Effect of Bone Marrow Mesenchymal Stem Cells Transduced with HSVtk Gene on Glioma Cell Lines

摘要: 目的探讨导入单纯疱疹病毒胸苷激酶(HSVtk)自杀基因的骨髓间充质干细胞(BMSC)对胶质瘤细胞的抗肿瘤作用。方法将导入HSVtk自杀基因的BMSC细胞分别与多种脑胶质瘤细胞株(人类和大鼠)共同培养后,采用MTT法观察其对胶质瘤细胞增殖的抑制作用。体外侵袭性实验观察胶质瘤细胞培养上清对BMSCtk的趋化作用。结果我们发现在更昔洛韦的存在下,人类BMSCtk细胞对胶质瘤细胞具有很显著的体外抗肿瘤活性。与新鲜培养基比较,从人类和大鼠胶质瘤细胞获得的条件培养基都可以增强人BMSC向胶质瘤细胞的迁移活性。结论导入HSVtk基因的BMSC在#x0201c;旁观者效应#x0201d;的介导下能够产生很强的抗胶质瘤效应。

Abstract: Objective To investigate the anti-tumor effect for human and rat glioma cells mediated by the bystander effect using bone marrow mesenchymal stem cells(BMSC) as a #x0201c;vehicle#x0201d;. Methods We valuated the anti-tumor effect mediated by the bystander effect using BMSCtk on glioma cells with a co-culture system. Results In vitroco-culture experiment showed a potent growth inhibition effects on glioma cells in the ganciclovir-containing medium. Migratory activity of the human BMSCs towards the tumor cells was enhanced by the conditioned medium derived from both human and rat glioma cells compared to fresh medium. Conclusion The bystander effect of BMSCtk cells is not cell type-specific,suggesting that the strategy could be quite feasible for clinical use.
