银杏内酯B对体外培养神经细胞延迟整流钾电流的作用

Effect of Ginkgolide B on Delayed Rectifier Potassium Current in SH-SY5Y Cells

摘要: 目的研究银杏内酯B对传代培养的SH-SY5Y细胞延迟整流钾电流(IK)的影响。方法传代培养SH-SY5Y细胞,用浓度为0.1、1.0和10.0#x003bc;mol/L的银杏内酯B灌流细胞,用全细胞膜片钳技术记录SH-SY5Y细胞IK。结果浓度为0.1、1.0和10.0#x003bc;mol/L的银杏苦内酯B在指令电压+60mV时分别使SH-SY5Y细胞IK峰密度值分别从(2.19#x000b1;0.45)pA/pF增大至(2.48#x000b1;0.37)pA/pF(P<0.05,n=5)、(2.97#x000b1;0.29)pA/pF(P<0.01,n=5)和(3.15#x000b1;0.48)pA/pF(P<0.01,n=5)。IK分别增大13.24%、35.62%和43.85%,具有浓度依赖性。结论银杏内酯B能增加IK,并且具有浓度依赖性。

Abstract: Objective To detect the effect of ginkgolide B (GB) on delayed rectifier potassium current in subcultured SH-SY5Y cells. Methods SH-SY5Y cells were cultured and perfused with GB 0.1,1.0,10.0 #x003bc;mol/L. Delayed rectifier potassium current (IK) was recorded by whole cell patch clamp technique. Results GB at concentrations of 0.1,1.0,and 10.0 #x003bc;mol/L increased IKdensity value of SH-SY5Y cells from (2.19#x000b1;0.45) pA/pF to (2.48#x000b1;0.37) pA/pF (P<0.05,n=5),(2.97#x000b1;0.29) pA/pF (P<0.01,n=5),and (3.15#x000b1;0.48) pA/pF (P<0.01,n=5),respectively. IKwasrespectively increased 13.24%,35.62% and 43.85% with concentration-dependent. Conclusion GB can significantly increase IKin a concentration-dependent manner.
