硫酸软硫骨素对3T3-L1细胞增殖和分化的影响

Effects of Chondroitin Sulfate on Proliferation and Differentiation of 3T3-L1 Cells

摘要: 目的研究硫酸软骨素(chondroitinsulfate,CS)对3T3-L1前脂肪细胞增殖及分化的影响,并对其机制进行初步探讨。方法噻唑蓝法观察0、5、10、25、50、100、200mg/L浓度的硫酸软骨素对3T3-L1细胞增殖能力的影响;3T3-L1细胞由3-异丁基-1-甲基黄嘌呤、胰岛素和地塞米松联合诱导分化,实验组加入硫酸软骨素干预,诱导分化第8天油红O染色观察脂肪细胞分化程度;荧光定量PCR检测过氧化物酶体增殖物激活受体#x003b3;(PPAR#x003b3;)、CCAAT增强子结合蛋白(C/EBP#x003b1;)和脂肪酸结合蛋白4(FABP4)mRNA表达水平;Westernblot检测脂肪细胞分化过程中PPAR#x003b3;、C/EBP#x003b1;和Smad3蛋白表达。结果硫酸软骨素对3T3-L1细胞的促增殖作用呈剂量和时间相关性,其中25mg/L的硫酸软骨素作用24h后对3T3-L1细胞的促增殖作用最大。硫酸软骨素能够抑制3T3-L1细胞成脂分化,减少脂滴聚集。并且硫酸软骨素能够下调PPAR#x003b3;、C/EBP#x003b1;FABP4mRNA,抑制PPAR#x003b3;、C/EBP#x003b1;、上调Smad3蛋白表达水平。结论在适当浓度范围内硫酸软骨素具有促进3T3-L1细胞增殖作用,并通过激活Smad3、下调PPAR#x003b3;和C/EBP#x003b1;抑制成脂。 t检验,P<0.05为差异有统计学意义。]]>

Abstract: Objective To study the effects of chondroitin sulfate(CS) on proliferation and differentiation of 3T3-L1 preadipocytes, and investigate the possible mechanism. Methods Chondroitin sulfate at different concentrations of 0, 5, 10, 25, 50, 100, 200 mg/L respectively were added during routine culture of 3T3-L1 preadipocytes. MTT assay was adopted to detect the proliferation. The cells were induced to differentiation by the cocktail medium containing 3-isobutyl-1-methylxanthine (IBMX) insulin and dexamethasone, and CS was added in the experiment group . On the 8th day of induction, the differentiation of 3T3-L1 cells was detected by oil red O staining. The expression of peroxisome proliferator-activated receptor #x003b3;(PPAR#x003b3;), CCAAT/enhancer-binding protein#x003b1;(C/EBP#x003b1;) and fatty acid binding protein 4 (FABP4) mRNA were determined with real-time fluorescence quantitative PCR (Q-PCR). Western blot was used to examine the protein level of PPAR#x003b3;, C/EBP#x003b1; and Smad3. Results Chondroitin sulfate promoted the proliferation of 3T3-L1 preadipocytes in a dose-and time-dependent manner, treatment with 25 mg/L for 24 h significantly increased the proliferation of 3T3-L1 preadipocytes. Chondroitin sulfate inhibited 3T3-L1 preadipocytes differentiation, and decreased lipid accumulation, the mRNA expression of PPAR#x003b3;, C/EBP#x003b1;, and FABP4, the protein of PPAR#x003b3;, C/EBP#x003b1; were all significantly reduced after CS treatment , while, the protein level of Smad3 was up-regulated. Conclusion Chondroitin sulfate promotes proliferation, and inhibits differentiation of 3T3-L1 cells, this effect may be related to down-regulation of PPAR#x003b3; and C/EBP#x003b1;, up-regulation of Smad3.
